GLUTEN ANALYSIS - ELISA Sandwich Analysis



Test purpose: this test allows the quantification of immunotoxic gluten in all species of wheat, barley, rye, including potential toxicity in oat.


Samples type: fresh or processed food products, homogeneous or heterogeneous, liquids or solids.


Limit of quantification: starting from 1.5 ppm gliadin.


Delivery term: 24-48 hours after sample arrival.


Technique description:

ELISA Sandwich is an immunological colorimetric technique that uses a monoclonal antibody both as a covering and as a detection antibody (peroxidase conjugated). This antibody recognizes gliadin epitopes, which are part of the gluten protein. During the test the samples change to a colour whose intensity is proportional to the toxic gluten concentration in the sample. The sample is quantified by comparison with a standard curve.


Control included in the test:

  • - Negative control: gluten-free maize flour.
  • - Positive control: maize bread with a known quantity of wheat gluten (55-65 ppm wheat gluten).

The sample, for more security, is analyzed in duplicate at two different dilutions.


Besides the ELISA Sandwich, this service also includes a parallel analysis by ELISA Competitive Sandwich with the G12 antibody (developed by the CSIC, University of Seville and Biomedal), which detects exclusively toxic gluten peptides. Therefore the analysis is double, offering the highest reliability.
